Can we have a post for the beginners for understanding NMDA nootropic action?

I'm new. AMPA receptors make a lot more sense than NMDA receptors when it comes to enhanced cognition, but they both seem crucial.

With NMDA receptors. Antagonists, agonists, and modulators all seem to have a complex role that I don't understand when it comes to cognition enhancement.

For instance Ketamine and PCP are an antagonistic yet so is Memantine and huperzine-A . Some cause Olney's Lesions and others are for Alzheimers.

Is there a NMDA explanation that is more straightforward out there?

Can somebody lay out NMDA for those in the dark like me?

What do you think this has to do with the CILTEP stack?

Yes, the traditional CILTEP stack is functioning on enhancing LTP from a different angle. However, NMDA function is key for LTP to occur. You can enhance the function of the AMPA, NMDA, and cAMP simultaneously. You just need to find the balance.

Activation of the NMDA receptors is necessary for LTP, but that transmission can be enhanced through the other pathways. The NMDA receptors must be activated by glutamate and glycine, and the cell must be depolarized to allow the magnesium blockage to be cleared. Only then can the activation by glutamate and glycine allow for calcium to enter the cell. This is mediated by the AMPA receptors. So the mechanisms are highly interdependent. Adding a positive allosteric modulator of the NMDA receptors along side your AMPAkine and cAMP supplementation is going to enhance LTP even further.
