Kohort study show large increase in hip fracture when high intake of b6 and b12!

I had this as a separate post but it was deleted by mods;

I'm taking a methylated b-complex with fairly high doses, and after reading up on b6 in anticipation of the new product release, I stumbled upon this. Most of us supplementing with a decent multi are easily over these levels.

Any thoughts? I was looking to bump my b6 to 75mg/day, with NDs new release but now I'm unsure.

You know what, after reading a bit more, I think there is something here. Vitamin B6 seems to modulate steroid hormones. Lower B6 levels actually corresponds to higher sensitivity to steroid hormones. This means people with higher levels of B6 are going to be less sensitive to estrogen in the body. In the study you linked, the hip fractures were mostly seen in low BMI post-menopausal women. Well skinny old women already make less estrogen than higher weight old ladies or younger women. Estrogen is a modulator of bone strength. So these skinny older women who naturally have lower estrogen are more sensitive to change in the body's sensitivity to estrogen. Higher levels of B6 in these women might push them over the edge with their low estrogen levels. It's also interesting when you think of how bodybuilders are using P5P to treat gynecomastia. P5P lowers prolactin, which will help with gyno, but it lowering the sensitivity to estrogen might be a factor in how well it works for gyno, too. Pretty interesting! It also seems to have a genetic component in relation to alkaline phosphatase.

The Alkaline Phosphatase \(ALPL\) Locus Is Associated with B6 Vitamer Levels in CSF and Plasma

The mechanism of mineralization and the role of alkaline phosphatase in health and disease

Genetic TNAP deficiency seems to be correlated with lower bone mineralization. Perhaps women with a genetic predisposition to lower TNAP activity are more susceptible to higher levels of B6 adversely affecting their bone strength. So the big question is: are you a frail post-menopausal woman? If no, I probably wouldn't worry about it. We are going to do some more research on all this before we finalize the B6 levels in our B complex, though. Thanks for bringing it up!
