Orexin-A and CNTF

Original post in this thread

So I've been trying out Ceretropic's Orexin-A for a week, still have about 3 weeks before I post a full review. As a person with Narcolepsy 2 however, this has been the single best substance I've ever tried, cutting my need for Amps/Armodafinil in half with better results. I had tried it for about 3 days when it barely arrived, however, I had a trip planned to the Dominican Republic so I left it at home, starting it up again just this week. I noticed that the great feeling I had during the first 3 days slowly starting waning by the end of my trip.

This leads me to a question: Can anybody (theoretically) think of a way to make this permanent? Would something that boosts CNTF help repopulate orexin neurons or orexin producing neurons in my hypothalamus?

Now, people who have Narcolepsy with Cataplexy, type 1, have an almost complete destruction of orexin in the hypothalamus. In the 2 autopsys done on people with Narcolepsy without Cataplexy, type 2, only a localized loss of orexin neurons was found:

https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2717206/ - Localized Loss of Hypocretin (Orexin) Cells in Narcolepsy Without Cataplexy

It also raises the question however, what's the fun…

What they were answering

So I'm still reading through the studies, gone through maybe half. My interest was piqued by the "Intranasal drug targeting of hypocretin-1 (orexin-A) to the central nervous system" study. They tracked orexin levels in different parts of the brain over the course of 120 minutes after administration. Is there any evidence that orexin-a administration would have an effect on orexin-producing neurons? Is there a peptide for that? (lol)

Orexin-A directly binds to the orexin receptor. That study was showing that intranasal administration was effective at increasing brain levels of Orexin-A, which then can go on to activate orexin receptors.

This study goes into pretty good detail on the receptor bindings: http://pharmrev.aspetjournals.org/content/61/2/162

Dipeptide analogs of piracetam, noopept, sulpiride,... exhibiting far superior potency and specificity to their putative mechanisms.

They are not talking about analogs of noopept. Noopept IS the analog they are discussing. The other compounds are not analogs of piracetam, either. They took inspiration from some of the functional groups, but they are V

IT'S HERE!!!! P21

Here's the thing, I want to avoid giving my own anecdotes for a bit, to prevent confirmation bias. I don't want people seeing how I react, then convincing themselves they react the same way. I want people to try it thems

Negative. This is a brand new peptide we developed with no human clinical research, or any research of any kind. It should be treated as a research compound, and should not be purchased based on anecdotal effect statemen
