So I've been trying out Ceretropic's Orexin-A for a week, still have about 3 weeks before I post a full review. As a person with Narcolepsy 2 however, this has been the single best substance I've ever tried, cutting my need for Amps/Armodafinil in half with better results. I had tried it for about 3 days when it barely arrived, however, I had a trip planned to the Dominican Republic so I left it at home, starting it up again just this week. I noticed that the great feeling I had during the first 3 days slowly starting waning by the end of my trip.

This leads me to a question: Can anybody (theoretically) think of a way to make this permanent? Would something that boosts CNTF help repopulate orexin neurons or orexin producing neurons in my hypothalamus?

Now, people who have Narcolepsy with Cataplexy, type 1, have an almost complete destruction of orexin in the hypothalamus. In the 2 autopsys done on people with Narcolepsy without Cataplexy, type 2, only a localized loss of orexin neurons was found:

I'll play armchair neurologist and posit that supplementing Orexin, if you can get it past the blood-brain barrier, would be picked up by other neurons as if the orexin-producing cells were still there.

The good thing is, it seems to cross the BBB quite easily in intranasal trials and to a lesser degree subq (http://www.jneurosci.org/content/jneuro/27/52/14239.full.pdf)

The downstream effects on supplemental Orexin does make sense, that supplementing makes other neurons act as if they were still there.

Please update us after your full month trial. If your experience isn't placebo and cuts your medication needs in half, this could be a huge benefit to narcolepsy patients.

I'm quite sure it isn't placebo, as I tend to nocebo my way through most supplements. The moment I realized that something was really different was when I woke up to use the bathroom one time after the second day of supplementation. Normally, I'll drunk stumble confusingly through the dark, not really knowing where I am or what I'm doing, and knock things over or bang my knee against the door on the way to the bathroom. However, I woke up, had my balance, and just knew I needed to use the bathroom and afterwards I would just go back to sleep as I hadn't heard my alarm yet. MOA seems to be related to modulation of glutamate, norepinephrine, melatonin, and possibly dopamine.
