Does this mean it has high affinity for the enzyme ? "Best substrate" is something I haven't encountered until now.
Also, mitochondria are incredibly interesting things (why do they have their own DNAs?) hehe .

Think of something being a good substrate, as being the same thing as having high affinity for a receptor, but instead of a receptor, it is for an enzyme. So something being one of the best substrates for Glutathione S-transferase P means that it binds to that enzyme better than most things. In the case of NAPQI, it binds to Glutathione S-transferase P, which then gets inactivated by glutathione. When your glutathione levels are low, NAPQI cannot be inactivated fast enough. That is why it is so toxic.
