Is tianeptine NMDA agnostic or antagnostic?

I would love some deeper explanation on this. Any directions you could point me towards?

I'm taking both concurrently.

So Tianeptine is modulating the current between the AMPA and NMDA receptors, right? The NMDA receptors are what opens to allow calcium into the neuron, and create an electrical potential, which allows signals to propagate across neurons. For the NMDA receptors to be able to open, the AMPA receptors have to activate, and depolarize the neuron. That allows the magnesium blockage in the NMDA channel to get out of the way, and allow calcium in. With Tianeptine modulating the current between the AMPA activation and NMDA depolarization, that is going to affect how Memantine binds. That is because Memantine binds to an open channel binding site on the NMDAR. So the magnesium blockage has to clear for that to happen. Since Tianeptine is altering the current from the AMPA receptors, the depolarization of the cell is going to be different.

I was postulating that since Tianeptine normalized those currents, that it would actually help Memantine's actions. They would both be working to prevent too much calcium from entering the cell from over-excitability/stress.

You need to start tapering off it now! You are not using it properly, and are going to cause yourself harm. Tianeptine is hard on the liver at those dosages. It is not meant to be taken at 650mg/day. Your usage is irresp
