I reckon its 5-HT2B activity is also responsible for MDMA's effects on cardiac tissue (specifically valves) that I have read about before. I find it very interesting that elimination of 5-HT2B receptors completely blocks the effects and serotonin release in mice. I figured it had minor effects at serotonin receptors, but to learn that it is essential for the effects of MDMA to manifest is fascinating.

Exactly! I knew of the 5-HT2B agonism, and the risks it posed for cardiac tissue. It's the same risk 6-APB has, albeit much lower. However, I did not make the connection to the 5-HT2B agonism being essential for the 5-HT release. It makes sense, though. 6-APB is more potent 5-HT releaser compared to MDMA, and also has higher 5-HT2B affinities.

This is kind of unfortunate, as the vasoconstriction is the worst part about 6-APB. However, the mechanism causing it looks to be central to it's 5-HT releasing effects. Some people were thinking of taking a 5-HT2B antagonist with it to prevent the vasoconstriction, but that looks like it would halt the good effects as well. Looks like L-arginine and taurine are the best defense at the moment.
