Nobody has measured the percentage in any of their lion's mane. The reference standards don't exist, nor do compendial methods. That was the whole issue with the fake dry lab Oriveda were using to prove that number. The numbers were not real. We have had to create our own reference standards, then validate them using quant NMR. That's what is happening in that 3rd link I posted. We are using our Buchi flash chromatography and prep HPLC to isolate and concentrate the various compounds, including hericenones. Once we concentrate enough, we then have to send off for validation to another lab that specializes in NMR (yes, even though it is a meme on other subs to say we don't use 3rd party labs, we send samples to 3rd party labs literally every week.). Then once it is all validated, we can use those standards to accurately assay for the content. We currently have developed UPLC methods that can separate all the erinacines and hericenones from each other, and can get a rough idea of content. However, we are still concentrating all the various hericenones and erinacines to get enough to use as reference standards for assay. This is why we are cultivating a bunch of our own lion's mane fruiting bodies using various strains. We need a lot of mushroom material to be able to get enough of all the erinacines and hericenones for reference standard creation, and not all strains make the same amount, or even all the different compounds. Right now our methods are best for identifying if there are erinacine or hericenones in a sample at all. If they are in there, we can make an estimation as to the content based on the peak size and our known response factor. However, we have a little more work before I will confidently say an exact percentage like we can now with ganoderic acids. I am hoping we will be able to do that in the next few months. It will all depend on the yields we get out of these fruiting body and liquid mycelium grows. When I say shit on the market is trash, it's because we don't need the reference standards to get an exact assay when the peaks are not even there. We've tested multiple products on the market making claims, and none of them have had any erinacines or hericenones at all. We don't need to run calibration curves with reference standards when there is no peak to even integrate.
In addition, as I was saying before, hericenones are only one part of the actives in the fruiting body. There is a whole other class of compounds in there that nobody is even talking about, and we believe those are the ones causing a lot of the neurogenic effects. We have concentrated reference standards of those compounds, and are in the process of publishing a paper on the whole thing. Also, if I gave you a % hericenone content, you would have no context for whether that is good or not. There has been ZERO validated chemistry on this from anyone so far, so you wouldn't know if a 10%, 1%, or a 0.1% number would be good. We have shown that erinacine-A specifically is active in the sub milligram amounts, so even in concentrations of 0.1% you can get active doses. The hericenones are going to be similar. Again, we have to validate all this chemistry before anyone can have even a cursory idea of what a "good" lion's mane extract should contain. We have the most cited lion's mane researcher in the world on our team, who has been working with our other PhD chemists in the lab for the past few years. Everyone cites his work on erinacine-A. Even the authors of those two Taiwanese studies on lion's man mycelium based their stuff off his work. We've been speaking to those researchers in Taiwan as well, and they even sent us some of the extract they used in those two studies. I've personally taken their extract multiple times, in addition to our own R&D batch of erinacine-A standardized liquid culture mycelium. Ironically, their sample doesn't contain erinacine-A... It contains another compound that looks similar to erinacine-A in certain tests, but is a completely different compound. It has an Rf value similar to erinacine-A(so, similar in polarity), but shows up as a different color than erinacine A when derivatized onto the TLC plate. In our flash experiments it shows up at 340nm, which means it has a chromophore similar to erinacine-A, but it is not eriancine-A. We ran mass spec experiments on it, and we are confident we know what it is now. It also explains why the Taiwanese researchers think they have erinacine-A when they don't. So we have some really big data that will rock the research world around lion's mane that will be coming out. The only two human clinical trials on lion's mane mycelium don't even contain the compound they claim to! Pretty crazy. I actually did not like their extract. It was really dysphoric for me. I could see how perhaps a clinically depressed person could have effects, though; so I don't dispute the results of their studies. However, the effects they are seeing are NOT from erinacine-A, as we have proven their extract doesn't contain it. I have also tried our own in-house R&D extract that did contain erinacine-A, and I loved it. A real erinacine-A standardized extract feels like no other lion's mane product. It just doesn't exist on the market yet.
So it is a lot more complicated than just giving you a % number for hericenones specifically. Anyone that has so far has lied to you. Even if I gave you that number, you wouldn't know how to contextualize it, and it would be missing another 2/3rds of the rest of the story. This is because there are two other classes of compounds in the fruiting bodies that give similar effects as well. I know I have said we will be releasing data soon for what feels like years now, but we are very very close on lion's mane. Real science just takes a lot longer than anyone realizes. It's taken years longer than I expected when I started this project, and a fuckload more money... These are not simple things. If they were, we would have been making erinacine and hericenone claims years ago, and selling more product. The fact that I refuse to make assay claims on these mushrooms till I am sure the science is correct and validated should say a lot. The fact that others have made those claims, then have been forced to remove them, should also say a lot. When we eventually make an assay claim, it will be accurate and validated with proper chemistry.
