Honestly I wouldn't be surprised if the pharmacokinetics also changed with the acetylation/amidation.
N-acetyl L-Tyrosine has different pharmacokinetics than L-Tyrosine. There are a lot of unknowns.

Of course the pharmacokinetics are different. That is the whole point! Did you mean to say pharmacodynamics? Pharmacokinetics is how the compound is absorbed, distributed, metabolized, and excreted. Acetylating and amidating of peptides is a very common modification to improve its pharmacokinetics.
