Interesting! That's crazy how many of these people skew or misconstrue their results just to make a quick buck. How do they sleep at night? I can't imagine becoming a scientist and dedicating my life to the pursuit of facts and objective truth, only to muddy the waters for everyone else.

I've tried comparing what you wrote to the metabolic pathway graphic, but I'm still a bit confused. I assumed the idea of NMN depleting methyl groups was plausible since NMN metabolizes into NR, which IIRC we have observed to increase the urinary excretion of methylated niacin. However, you noted that

NMN can then go multiple ways. It can bioconvert to NR, which then is uptaked into cells, converted back into NMN, then converted to NAD+, or it can proceed directly to increasing NAD+.

So my questions would be,

* What's the point of NMN's bioconversion to NR if it is only to be reconverted? Maybe I just don't understand how vitamins work on a molecular level. Do they exert some action while remaining unchanged within the cell, kind of like an enzyme? (I definitely feel like this is a stupid question, LMAO)
* Does this mean that NMN's impact on niacin status is minimal? I know I will be taking the ND NMN due to the NAD+/longevity benefits, but I'm also looking to improve my current niacin status.

In large houses with lots of money? I mean, that's disheartening to hear, but that's the truth. There is a FUCKLOAD of money being made on some of this shit. Patented supplement ingredients make a LOT of money. When there is a lot of money to be made, there is a lot of money in skewing the public discourse in your direction. I can make science say anything I want. That's what most people don't realize. You can misuse science to confuse people and make it look like something is true that really isn't. Even I did not know the scope of that till I built my own lab, and hired really really intelligent scientists. I think this is the most damaging thing facing our industry right now, in fact. People naturally distrust anecdotes. They know people lie in marketing. They know companies cut corners to make more profit. They don't know that science can be done in such a way that makes it look legit, but is actually false or misleading. The last thing we need right now is to give people a reason to distrust science! There's enough science denial already. We need to make sure that no extra fuel is added to the fire; legitimate fuel, as well...

This is why I am so passionate about valid chemistry and valid science! I am trying to make people care about something that matters more than anyone thinks! You HAVE TO be able to trust numbers you are seeing, or people start to distrust science as a whole. Showing a fake number for erinacines and hericenones has wider effects than just people being misled on lion's mane specifically. This is a fight for truth. It's a fight for trust in science. We have the responsibility to ensure we verify and validate this science before we attempt to position ourselves as an authority. This is also why things take us a lot longer these days. We are not being lazy and procrastinating. I literally have teams of people in the background doing detailed chemistry and scientific research to validate things to an extremely high level. That way when I come here, I can be damn sure what I am saying is based in real validated science. It's something I am trying to get everyone here to care about as much as I do, because this affects everyone.

The methyl donor thing only comes into play if niacinamide levels increase significantly. The thing with NMN is that it is rapidly absorbed in the small intestine. The people promoting the methyl donor thing are talking about supplementing exogenous niacinamide instead of NR and NMN. That will lead to high levels of methylated niacin in the urine. NMN has its own transporter in the small intestine that NR can't use, though. So we are talking about getting whole NMN into systemic circulation. NMN is not going to increase niacinamide levels like an oral dose of niacinamide would, or even an oral dose of NR. NMN can convert to NR and then right back to NMN inside of cells. NMN can also go directly into some cells and work without converting to NR first. Remember, taking NR orally is going to be very different than NMN quickly converting to NR to enter a cell, then converting back. Also, let's look into their theory a bit more, as it kind of seems dangerous what they are promoting.

Niacin is likely more therapeutic since it produces higher NAD levels than niacinamide, while also activating GPR109a, followed by activation of PPAR-γ.

So if you were going to go the route they are suggesting, taking niacin would be better than taking niacinamide, since it leads to greater increases in NAD+ and other beneficial effects. Then let's hear what they have to say about the methyl donor and niacinamide thing.

Then the author discusses how niacin would increase NAD and niacinamide levels, leading to the methylation of niacinamide to 1-meth-ylniacinamide (MNA) in the brain. This might potentially be harmful as superoxides formed by MNA, via complex 1 of the electron transport chain, can destroy complex 1 subunits and directly or indirectly damage mitochondrial DNA. Furthermore, this mitochondrial damage might be accelerated by hereditary or environmental factors resulting in neuronal death.

So there is evidence that taking niacinamide with methyl donors could damage mitochondrial DNA. It's something I have to research more.

Some cells don't have the NMN transporter on them, or at least we have not found it yet. So the conversion to NR allows them to pass the cell membrane, where they are converted back to NMN as needed by eukaryotic nicotinamide riboside kinase (Nrk1 and Nrk2).

Well remember that NAD+ can convert back into niacinamide, which can then convert back to niacin through the nicotinamidase pncA pathway. So yes, NMN should lead to an increase in niacin as well. However, the amount will be controlled by three rate-limiting enzymes.

You should mention NMN's exclusive absorption within the small intestine on the product page! Right now it only mentions the enteric coating being added for increased stability.
