Redosing MDMA or use Amphetamine

Original post in this thread

Hey /r/drugs,

I know that redosing MDMA increasing the neurotoxicity, so I am wondering if using Amphetamines instead of redosing would be less neurotoxic. Also later redoses mostly bring out speed-like effects, so I am assuming it would be better to just use actual speed.

The Amphetamine in question is pharmaceutical D-amphetamine (Dexedrine). I am going to a big EDM show in around 2 months and probably an after party, so I want to be able to keep on partying.

Thanks :)

The most popular theory at the moment is that the metabolites from mdma cause the neurotoxicty.

I wouldn't say most popular, but it is a theory. There are multiple hepatic metabolites of MDMA that can be toxic. In fact, around 10% of your dose becomes MDA under normal conditions, which can then be further broken down. However, this theory cannot explain why those with a CYP2D6 enzyme deficiency do not show decreased neurotoxicity. In fact, they show increased serum levels of MDMA and slightly higher neurotoxicity in some cases. It's been postulated that MDMA itself can be broken down by MAO-B in serotonin neurons to toxic substances. Another theory is that dopamine is the culprit being broken down. This makes sense why MDA would be more neurotoxic, since it has a higher DA affinity. It also make more sense for why methamphetamine is more neurotoxic. My theory is that it's a combination of everything. We've proven all these substances can be neurotoxic under the right conditions. So the right conditions are when your body temperature rises and makes it harder for antioxidants to protect you from these reactive oxygen species. Why does it just have to be one substance causing toxicity?

The metabolism characteristics of the drug mean that the first dose inhibits the main metabolic pathway, CYP2D6. This means that any subsequent doses are more likely to use the secondary metabolic pathway, CYP3A4. The CY
