Protein aggregates occur in a lot of neurodegenerative diseases. I suspect they're usually caused by metabolic problems and targeting them directly will never be effective. A common feature of these diseases is a disruption of NAD+ synthesis enzymes, particularly NMNAT2, which is why SARM1 inhibitors look promising. HSP90 inhibitors are also effective in a variety of disease models, which is why I keep hoping you guys will come out with a standardized thunder god vine root (celastrol) extract.

That would make a hell of a lot more sense than the plaques being the cause of the damage. What we are likely going to find is there are multiple mechanisms that play into the progression of the disease, and targeting a single one is not going to be effective.

The issue for me there is the toxicity and risk of adverse events that we still don't fully understand.
