A couple of days ago, I went out and partied the night before and important meeting. I drank a little (lot) too much and woke up with a pretty bad hangover the next day.

In a hail mary attempt to not feel like shit and be able to make the meeting, I randomly decided to take 25mg of MK 677 that I had leftover from an experiment. It was about a month since I had last taken it.

It worked! My nausea and sickness was quickly replaced with an extreme hunger. Rather than nibbling on a piece of toast with some Gatorade, as I normally would when I'm hungover, I smashed 5 eggs, 2 pieces of toast, and 3 glasses of orange juice.

I still had a minor headache for the rest of the day, but nothing even remotely close to how shitty I felt before taking MK677.

So perhaps the peripheral increase in GABA is causing a systemic lowering of GABA release in the brain. I would be curious to see how you react to Phenibut or Baclofen, as those pass the BBB a lot better.

That was my theory on why phenibut has been reported to increase tolerance much faster than baclofen. Maybe phenibut is mostly peripheral and there is a systemic downregulation of GAD65/67, GAT etc?

I wonder what would happen if someone made a peripheral GABA antagonist that doesn't cross the BBB? Would it upregulate GAT etc in the brain?

I also know that there are ~17 different gaba A receptor subtypes that aren't really well studied. But there are only a few GABA B receptors. Maybe different drugs activate these unstudied GABA A receptors and cause various homeostatic mechanisms to trigger.

Could be. I think GABA needs to be studied more than it has been. We know so little about it compared to other systems. The newer Russian nootropics are getting into it a bit. However, we need more research interest to get a clearer picture of all the interactions.
