Having had recent success from supplementing methylfolate and methylcobalamin (*) and in turn ordering a test from 23andMe to more diligently iron out a supplement regimen, I'd like to inquire about a few things:
- For those who have had success (in a nootropic sense) from testing for and treating methylation issues, what's your story and regimen? What benefits and/or costs have you experienced?
- It's occasionally brought up that methylation treatments are not backed up by sufficient evidence. From the perspective of someone well-versed in the area, what's your take? Is it akin to something like common nutritional deficiencies, where the symptoms are well-known and blood tests readily available, but literature is lacking on treating deficient individuals back to baseline levels and recording the results? Or is something more grave at play?
- Finally, aside from Genetic Genie, 23andYou, and Promethease, are there any other 3rd party venues worth checking out?
(*) Background context: Tested deficient for B-12 over a year ago at a time when I was consistently eating heavy amounts of animal products. Started receiving injections and taking sublingual M-B12 until mostly ameliorat…

I'm highly skeptical about methylation claims. If methylation was that important as they claim, supplementation of methylation end products would be at least as effective as expensive methyl donors, if not more.
Such end products include:
- Creatine
- Choline
- Carnitine
- Coenzyme Q10
- Melatonin
That <5 mg Methylfolate or Methylcobalamin won't make a difference, only in the thickness of your wallet.
Edit: My point is that supplementation of methylation end products free up methyl groups to be used for other purposes. Geez impulsive downvoters who are incapable of thinking.

I am not sure you realize how complex the methylation cycle is, and all of the end products it affects. Serotonin, norepiniephrine, dopamine, histamine, glutathione, etc. are all heavily dependent on the methylation cycle. You cannot just supplement end products for everything.
Addressing my methylation issues has changed my life, and I have spoken to many other people that are in the same boat as I am. Methyl-B12, TMG, and SAMe are extremely effective in increasing my energy and mood levels. I am still not getting anywhere on the histamine front, but that is much less studied than the others. But it certainly is not just making my wallet thinner.
