This is probably going to be the first in a series of discussions I start about MDMA. There's just too much information for one post. Therefore, I am going to start with one that is very interesting to me: MDMA's metabolites and their role in neurotoxicity. I pre-appologise for the length and terminology used.

First off, let's discuss how MDMA is metabolized. The human cytochrome CYP450 is responsible for the metabolism of MDMA. The primary enzyme responsible is CYP2D6, using O-demethylation. This process adds two hydrogen atoms to the two open oxygen atoms in MDMA to create HHMA. Let's look at the structure for a minute.

MDMA is 3,4-methylenedioxy-N-methylamphetamine

HHMA is 3,4-dihydroxy-N-methylamphetamine

So your CYP2D6 enzyme added two hydrogen atoms to the methylenedioxy structure to create a dihydroxy structure. Once it's been o-demethylated to HHMA, it is no longer active like MDMA is. HHMA can then be 0-methylated further to HMMA, or 4-hydroxy-3-methoxy-N-methylamphetamine. Here is an image to help you visualize this process.

Maybe this is a weird question but does all of this preparation enhance the experience during? Or does it just ensure (or greatly reduce) the chance of a bad crash? Personally MDMA (~115mg) for me only lasts around 4 hours, I think I'd spend 90% of my time just thinking about when to take the next supplement.

Absolutely, yes! I've taken it where my downward rolls were pure joy and my upward rolls I was literally shaking from euphoria. I had no jaw clenching, and no urinary retention. That night I fell right asleep, and woke up feeling like a million bucks. Then I had a 2 week long afterglow. I have not had a single crash from MDMA, or any adverse side effects, using my current supplementation. I re-dose at between 1 and 1.5 hours after drop. Usually it's about a 6 hour roll for me. After that, I am content to crawl under the sheets and sleep. Then the next day, all my meals and beer are the best things I've ever tasted. The next day is a treat just like the night of.

I don't have an issue remembering to take anything. The during roll supplements are just alpha lipoic acid every hour or so, magnesium glycinate halfway through, and emergen-c packets and grapefruit juice sometime through your roll. Then you go back to your house/hotel room/tent and just take what's preplanned in your pill container. Have it all labeled and ready ahead of time so you don't have to think about it before bed.

Keep in mind that I get really pure MDMA crystal, and I dose exactly. Pills are a total shot in the dark these days. I still get good pills too, but I have a good contact. The street stuff is scary.
