OptiNAD+ and tinnitus

Last week there was a OptiNAD+ Reviews Thread in where 4 different users experienced tinnitus after dosing OptiNAD+. This is pretty concerning, especially considering one of the users contacted ND support and they reportedly refused to help, speculating it was due to potential legal issues. I really hope that's not the case, ND staff always discuss all the positive impacts of their supplements, but surly they wouldn't ignore a side effect that multiple customers are experiencing. I'm posting this thread in order to get a bit more visibility and discussion around the issue. Hopefully someone from ND can jump in and get involved as well.

For those who haven't experienced tinnitus consider yourself lucky, it is not a pleasant condition.

We got one email regarding tinnitus and OptiNAD+, and our support team responded right away. The person was speculating that it might be from depletion of methyl groups, and Rachael from our support team told him that is what the trimethylglycine was in there for. She also said that often times reactions can be very complex, and it is difficult or impossible for us to speculate as to why one person might react one way to something. She also explained that we are limited in our capacity to respond by the FDA as well, as we are not doctors. We cannot attempt to fulfill the role of a physician by diagnosing, preventing, treating, or curing a disease. The person who emailed us said he was going to go see his doctor about it. That's where it was left.

To start, tinnitus is a poorly understood issue, and likely a very complex mechanism. I have bad tinnitus from not protecting my ears from sound systems in clubs that were running them too high, and a firework that went off next to my head. It's constant for me, and something I will just have to live with the rest of my life. My father told me when I was young to protect my ears, but we are all idiots when we are young, so I didn't really listen. I wish I had. Anyone here reading this: PROTECT YOUR EARS! So that is a pretty complex discussion. However, they did use flavoprotein autofluorescence (FA) to measure changes in the redox state of NADH and NAD+ to detect hyperactivity in the circuits related to tinnitus. They are saying it is related to a lack of glycine inhibition and decreased GABAergic inhibition. So what in OptiNAD+ could be leading to some people noticing tinnitus more? Perhaps quercetin?

Quercetin antagonism of GABAAρ1 receptors is prevented by ascorbic acid through a redox-independent mechanism

So that is saying that quercetin is a GABA-Aρ1 antagonist, which can be counteracted by vitamin C. Well I take Reacta-C every morning with my OptiNAD+, which could be why I don't notice any issues. Also, there is another elephant in the room: COVID. There is a clear link between COVID-19 and tinnitus. There is also a link between COVID vaccines and tinnitus. I can say for absolute certainty that I had higher tinnitus after every COVID vaccine dose, and I had tinnitus increase from actually getting COVID.

I am not the only one that has had their tinnitus go crazy with COVID and the vaccines for it. I would assume everyone here has either had COVID, the vaccines for it, or both recently. This creates a massive confounding variable to this whole situation. So is it the GABA effects from the quercetin? Could it be the trimethylglycine reducing histamine levels in the brain, which could set it off in some people? Is it just increased neuronal excitability from increasing NAD+? Some people have reduction in their tinnitus from using ginkgo, which is a negative allosteric modulator of GABA. However, other's have their tinnitus set off by it. Some people have improvements in their tinnitus from baclofen. Other people have it set off. Unfortunately we have waded into a very complex issue that is going to take more than our speculation to solve.
