Interesting that this more potent analog has a toxicity profile, while piracetam and aniracetam were shown to be unable to produce any toxic effects (iirc, an LD1 couldn't even be found in rodents, and they gave up trying. the lack of any toxic effects was the [hilarious] reasoning behind the FDA ruling against classifying piracetam as a drug).

Well the molecule is significantly different. The toxicity arises from one of it's metabolites. See this figure. M-10 is the one they believe is the cause of the toxicity. Animals metabolize nefiracetam differently than we do, so humans may not have the same issue.
