If I drink Friday night and Saturday all day, should I take AD after each night I’m done drinking? Or only after I’m done completely Saturday night?

When you are done drinking for the day/night. So I would take it when you are done drinking Friday night. Then I would take it when you are done drinking Saturday. The reason we say not to take it before drinking is that it will make you feel less drunk, which might make you drink more than you would have. There's no harm in taking it before/during, as long as it doesn't lead to you drinking more alcohol than you would have without it. That's the reason we tell people to take it after drinking.
