Water for reconstituting peptides into nasal sprays?

Original post in this thread

Hey guys,

I'm awaiting the arrival of a few vials of peptides (Semax, Selank, P21) that I'll be reconstituting into nasal sprays, but I have a few questions about which type of water to use.

I've done some research on this subject and the consensus seems to be that high quality deionized water (ASTM Type 1/2) should be the best option due to being the most stable. If ASTM Type 1/2 is unavailable, the next best thing according to the recommendations seems to be Bacteriostatic Water (BW), while distilled/sterile water seems be recommended against.

Neither ASTM Type 1/2 or BW is easily available in my country though and I haven't come across a good source elsewhere in Europe either, which got me wondering why BW would be any better than something like Sterile Water For Injection (SWFI)? To my knowledge, neither BW or SWFI is deionized, in which case wouldn't they both be equally stable? I don't think SWFI has any bacteriostatic properties, but neither does ASTM Type 1/2 to my knowledge, which seems to suggest that it shouldn't be a concern for nasal sprays anyway? Am I correct or misinformed in these assumptions?

Also, approximately how long would the reconstituted peptides be go…

I truly appreciate the feedback mate, the problem is just that it doesn’t quite add up and my questions outlining the inconsistencies of the information are being ignored:

---

1: How is ASTM Type 1/2 generally recommended as the overall best and most stable option if bacteriostatic properties are important for stability/safety?

You personally haven’t suggested using ASTM Type 1/2, so you might not share the opinion that it’s suitable, but I’ve read a bunch of recommendations on this and Ceretropic supposedly use this for their pre-made sprays. The reasoning is that ions will catalyse the breakdown of peptide bonds, so the lack of them in deionized water will keep the solution stable for longer.

/u/Majalisk suggested that ASTM Type 1/2 might actually be bacteriostatic because it might kill organisms by ripping them of ions, but it seems to be more of a theory than a fact.

2: If bacteriostatic properties are indeed not important for stability/safety, as suggested by question #1 unless more conclusive information on the bacteriostatic properties of ASTM Type 1/2 is presented or until the use of ASTM Type 1/2 is found unsuitable because of the lack of bacteriostatic properties, then why would Bacteriostatic Water be any better than Sterile Water For Injection?

3: Are medically used nasal sprays like saline and decongestants bacteriostatic? This could possibly help answer whe…

18 megaohm DI water (type 1) will be very hard for microbes to grow in, if not impossible. That doesn't mean they are not in there, though. They just won't go into the growth stage. They could start growing once they reach more nutrients, though. This is why you need UV treated type 1 DI water. That uses a UV step to ensure any microbes left are killed. Then it will be very unlikely anything would grow in it, even without an antimicrobial in there. That being said, it's better to be safe than sorry in this aspect, since you might be injecting it or putting it into a nasal spray, which will bypass much of your immune system. So using some antimicrobial is a good idea.
