I take it that means they're really dangerous to take in combination? If it's getting released but not taken back up, that sounds like your brain would be awash in it and unable to cope.

You mean cocaine and amphetamine? Actually cocaine will stop the amphetamine induced release, since it blocks the DAT and SERT. Amphetamine is already blocking the re-uptake, so the cocaine will just be counterproductive. There is also some evidence that re-uptake inhibitors can protect from amphetamine neurotoxicity, since it prevents the oxidative substances from being uptaked into the neuron.
Most of the issue with amphetamine is the fact that it inhibits the VMAT in the neuron. This allows dopamine to be broken down by MAO outside of the vesicles. This can damage the neuron. Reuptake inhibitors like cocaine and methylphenidate will actually block the DA from entering the neuron. This could actually provide short term mitigation of oxidative stress. I would not suggest it, though. Selective MAOIs like selegiline are much better suited for this.
